BACKGROUND

Murray Valley encephalitis virus (MVEV) is a mosquito‑borne flavivirus endemic to Australia and parts of Papua New Guinea that can cause severe, sometimes fatal, encephalitis in humans during episodic outbreaks (Mackenzie et al., 2017). The non‑structural protein 1 (NS1) of flaviviruses is a secreted glycoprotein that plays roles in viral replication and immune evasion and is a major target of the humoral immune response, making it an important antigen for both diagnostics and vaccine research (Slon Campos et al., 2018).

Recombinant NS1 protein from MVEV can be expressed and purified to provide a well‑defined antigen that does not contain the virion surface envelope (E) protein, thereby avoiding many of the cross‑reactive epitopes associated with E‑directed antibodies to multiple flaviviruses (Young et al., 2000; Henchal & Putnak, 1990). Such recombinant NS1 preparations are suitable for use in serological assays aimed at detecting NS1‑specific antibodies and for experimental immunization studies assessing the contribution of NS1 to protection and pathogenesis in MVEV infection models (Slon Campos et al., 2018; Mackenzie et al., 2017).
