BACKGROUND

Human epididymis protein 4 (HE4), also known as WFDC2, is a secreted glycoprotein first identified as overexpressed in serous and endometrioid ovarian carcinomas (Drapkin et al., 2005). HE4 is clinically useful as a serum biomarker in ovarian cancer, including for diagnosis and monitoring, and recombinant HE4 provides a defined antigen for studying its biochemical properties and for supporting assay development (Moore et al., 2008; Sokoll et al., 2014).

Recombinant HE4 has been produced in both mammalian and yeast expression systems and shown to be secreted and glycosylated, with glycosylation influencing its biochemical and biological activity (Sokoll et al., 2014). In addition to its diagnostic value, HE4 has been implicated in ovarian cancer progression, and recombinant HE4 has been used in studies exploring tumour-related functions such as proliferation and chemoresistance (Giamougiannis et al., 2023).
